چکیده
Abstract Therapeutic radiation can result in substantially different survival outcomes for patients with non-small cell lung cancer (NSCLC). Measures identification of who benefit most throughout radiotherapy remain limited. In this retrospective study, analysis was performed based on a discovery cohort from TCGA and validation three independent hospitals. Tumor mutational burden (TMB) chromosomal aneuploidy (ANE) were derived the whole exome sequencing (WES) data treatment-naïve tumors. Integrated risk scores TMB ANE by multivariate Cox proportional hazards model. reveal that are associated positively negatively, respectively, radiotherapy. Additionally, synergistically predictive significance these two genomic alterations, differing responders non-responders to is identified. These biomarkers may have clinical potential improve personalized treatment management rationally identifying highly likely therapeutic NSCLC.

متن کاملTGFβ1 Polymorphisms Predict Distant Metastasis–Free Survival in Patients with Inoperable Non-Small-Cell Lung Cancer after Definitive Radiotherapy
PURPOSE Transforming growth factor (TGF) -β1 signaling is involved in cancer-cell metastasis. We investigated whether single nucleotide polymorphisms (SNPs) at TGFβ1 were associated with overall survival (OS) and distant metastasis-free survival (DMFS) in patients with non-small cell lung cancer (NSCLC) treated with definitive radiotherapy, with or without chemotherapy. METHODS We genotyped T...
متن کاملQuantification of circulating cell-free DNA to predict patient survival in non-small-cell lung cancer
We used computed tomography (CT) to explore the prognostic value of cell-free (cf) DNA quantification and its predictive efficacy over time after chemotherapy in non-small-cell lung cancer (NSCLC) patients. In total, 177 NSCLC patients were enrolled in a prospective biomarker trial. Consecutive paired blood collection was performed to determine cfDNA concentrations at baseline CT and throughout...
متن کاملInterleukin-10 Haplotype May Predict Survival and Relapse in Resected Non-Small Cell Lung Cancer
IL-10 is associated with tumor malignancy via immune escape. We hypothesized that IL-10 haplotypes categorized by IL-10 promoter polymorphisms at -1082A>G, -819C>T, and -592C>A might influence IL-10 expression and give rise to non-small cell lung cancer (NSCLC) patients with poor outcomes and relapse. We collected adjacent normal tissues from 385 NSCLC patients to determine IL-10 haplotypes by ...
متن کاملVinorelbine-based chemo-radiotherapy in non-small cell lung cancer.
AIMS AND BACKGROUND Concomitant radio-chemotherapy improves survival of patients with locally advanced non-small cell lung cancer, with a better local-regional control. METHODS AND STUDY DESIGN We report our experience with vinorelbine-based chemotherapy in neoadjuvant and radical settings in 43 patients. Regimens consisted of cisplatin plus vinorelbine in 74.4% patients and carboplatin plus ...
